
The number of calcified nodules in the CCC+BMP7 group was significantly higher than that in other groups. The osteogenesis related genes, COLI, and integrin-Î☒, -α5, and -Î☡, were expressed significantly higher in CCC+BMP7 BMSCs. ALP activity and osteaocalcin content were significantly increased in CCC+BMP7 BMSCs. CCC+BMP7 BMSCs showed higher proliferation rate.

Cell proliferation, alkaline phosphatase (ALP) activity, osteogenesis related gene expression, osteocalcin levels, and calcified nodules were quantified and compared between four groups: untreated controls, BMSCs cultured with CCC complex medium, BMP7-overexpressing BMSCs, and BMP7-overexpressing BMSCs cultured with CCC complex medium (CCC+BMP7). BMP7-overexpressing BMSCs were generated by lentiviral-mediated gene transduction. In conclusion, simultaneous presence of HA and CCC contributed to the stem cell activity and epidermal differentiation in the reconstruction of SE. It showed that epidermal differentiation was significantly improved by addition of CCC.

Furthermore, filaggrin expression was much increased when CCC was added. These finding showed that addition of HA and/or CCC may affect the stem cell activity in the reconstruction of skin. Increased staining of integrin Î☖ and Î☡ was variably observed when HA and/or CCC was added to make dermal substitute. But, immunohistochemical staining showed that p63 was significantly increased by addition of HA and/or CCC. Results showed similar morphologic findings when hyaluronic acid (HA) and/or CCC was added. Four different models were constructed according to different composition of dermal substitute.

The aim of this study was to investigate the effects of Cervi cornus Colla (CCC) on the stem cell activity and epidermal differentiation in the reconstruction of skin equivalent. In the reconstruction of skin equivalents (SEs), keratinocyte differentiation is important because epidermal differentiation is closely related with barrier function. The cornelian cherry dogwood is the first dogwood to flower and is one of the first plants to flower in the Midwest in early spring.

The gray-brown to rich-brown exfoliating, flaky bark, is very attractive. Fall color can be reddish to purple, depending on the year. Bright red fruit that ripens in midsummer is edible but extremely tart. Small yellow flowers appear in March, and during a cool spring, they'll last for weeks. It is available both as a single- or multi-stemmed plant and can be a nice specimen tree if its lower branches are pruned. Cornelian cherry dogwood offers some of the earliest spring flowers, vibrantly colored summer fruits, and exfoliating bark that adds rich visual interest to winter gardens.
